The use of micro-organisms or their enzymes in the industry is ubiquitous and an essential part of a sustainable biobased economy. Part of indusrial biotechnology is the recombinant production of biomolecules, such as industrial enzymes, therapeutic proteins and bioactive molecules. A wide variety of host cells is available for recombinant production, but no single host is able to produce every desired biomolecule to commerciallevels. An interesting host cell, because of, inter alia, its high secrection capacity, is Streptomyces lividans.

This PhD aims tofurther improve S. lividans as an industrial host cell by targeted genetic modifications. Hereto, different bioreactor experiments are performed in which both the gene expression and the metabolic fluxes in S. lividans are mapped. The gene expression and flux data will be integrated into a mathematical model that will serve as a base for geneticstrain improvement.

Specifically, the recombinant production of cellulase and xyloglucanase will be studied. These are both industrial enzymes with applications in the paper, textile and biofuel industry. Furthermore, the effects of a genome reduction, in which the genes for several non-essential metabolic pathways are deleted, will be studied. The genome reduced strain will be used to study the expression of cellulase and rebeccamycin, the latter of which is a metabolite with significant antitumor properties.
